Artificial metalloproteins based on heme acquisition protein HasA
Episode 5: Complexation of Pseudomonas aeruginosa HasA with 5-oxaporphyrinium cation complex
So far, we have succeeded in conjugating various metal porphyrin complexes, mainly iron-DPP complexes, to HasA. Still, in collaboration with Prof. Shinokubo’s group (Nagoya University), we have investigated conjugation with even more unique metal complexes.
The target was a cobalt complex with a 5-oxaporphyrinium cation (Co-OEOP), which has a structure that is seen as an intermediate in the decomposition of heme in Pseudomonas aeruginosa and has a skeleton in which an oxygen atom is inserted into a part of the porphyrin ring. As expected, HasA formed a stable complex with this porphyrinium cation. Compared with cobalt-octaethylporphyrin (Co-OEP), which has a normal porphyrin skeleton in which the oxygen atom of Co-OEOP is replaced by carbon, HasA showed a high growth inhibitory effect against Pseudomonas aeruginosa and also showed a growth inhibitory effect against multidrug-resistant Pseudomonas aeruginosa.
